SLU-PP-332

ERR agonist (exercise mimetic) · Also known as SLU-PP332, Exercise Mimetic Compound 332

What is SLU-PP-332?

A small molecule that activates estrogen-related receptors (ERRs) to mimic the molecular effects of exercise, including increased endurance, fat burning, and muscle fiber remodeling.

SLU-PP-332 activates all three estrogen-related receptors (ERRalpha, ERRbeta, ERRgamma), which are key regulators of energy metabolism normally activated by exercise. In mice, it increased running endurance by 50% and induced a shift toward fatigue-resistant muscle fibers.

Key takeaway: SLU-PP-332 is an 'exercise in a pill' compound that mimics exercise adaptations at the molecular level, but remains in early preclinical research.

How it works

Exercise activates estrogen-related receptors (ERRs), which turn on genes involved in mitochondrial biogenesis, fatty acid oxidation, and muscle fiber type switching. SLU-PP-332 directly activates these same receptors without the need for physical exertion.

In mice, this resulted in a shift from fast-twitch to slow-twitch (fatigue-resistant) muscle fibers, increased mitochondrial density, and enhanced fat oxidation. Treated mice ran 50% longer on a treadmill test and resisted weight gain on a high-fat diet.
